Was in Santiago in late August and stayed downtown at the Hotel Plaza San Francisco. This is a 4/5 star hotel and I think the rate was $80 or so a night for two (web site seems to indicate much more - that is likely their rack rate). This hotel is in the historic district and you can walk to see many of the local sights. If you go to the website and click on "services" and "tours," you will find the descriptions of several tours that will take you to the wine country. Several tour companies operate out of the plaza next to the hotel (home of the oldest church in the city). We were on a skiing trip in the Andes and drove through the wine country to get to the ski resort (which is coincidentally on the road to Argentina). Another ideas would be to visit the wineries and then stay at the ski resort as a mid-way point to Argentina. http://www.skiportillo.com will give you some info. Also, if you stay at Portillo, they have a fantastic wine list and they have verticals of Don Melchor (for as long as Conha y Toro have been making a single vineyard bottling).
Have fun.

I cannot give you a recommendation for a town or hotel, etc. But we were in Chile in February and spent several days in Santiago prior to boarding our cruise ship. May I recommend finding the Santa Rita winery in the Maipo Valley. Their better wines are superb. Also, look up a light white wine known as "30 Sur" (a latitude, I'm sure). Lightly chilled it's very clean tasting and refreshing. The waiter in the cocktail lounge in our hotel suggested it and we liked it so well we went to a Super Mercado and bought several bottles to take with us on the ship.Great country. Enjoy your trip.
santiago and maipo are definitely on the good list... wonder if the '30 sur' is available here?? santa rita's are, but usually it's the everyday stuff i see.
